What are benefits of storage at -152C?

What are disadvantages of storage in liquid nitrogen vapor?

Mechanical cryogenic freezers offer significant benefits over conventional Liquid Nitrogen techniques. Liquid nitrogen storage systems are vacuum-insulated chambers filled with LN2 and periodically refilled as the LN2 evaporates.

This method, however, poses a number of concerns that may affect cell viability including significant temperature variations throughout the chamber, enough to allow a significant portion of cell load to be subjected to insufficient temperatures above the critical -130C recrystallization point of water.

In addition, mycoplasma cell and tissue contamination, plus a higher potential for cross-contamination, is due to brittleness of vials, bags or other containers which can break when exposed to liquid nitrogen at -196 C. There are always dangers of sudden liquid eruptions and the hazards involved in the handling of liquid nitrogen. Costs of maintaining liquid nitrogen supplies inflate operational costs as well.
